EPS是微生物聚集体的重要组成部分,提取和分析好氧颗粒污泥的EPS利于深入研究这一新兴微生物聚集体。采用加热、超声、高速离心和加碱等5种方法提取好氧颗粒污泥的EPS,并分析其主要成分。结果表明,匀浆预处理是提取颗粒污泥EPS首要步骤,超声和加热分别适合EPS定性与定量分析,在35 W、超声4 min的条件下,活性污泥与好氧颗粒污泥EPS提取液中TOC产量分别为105.3 mg/g VSS和96.5 mg/g VSS;加热法(80 ℃,60 min)提取的EPS产量略高于超声法,对于活性污泥和好氧颗粒污泥,TOC含量分别为142.6 mg/g VSS和153.2 mg/g VSS;蛋白质是活性污泥和好氧颗粒污泥EPS中比例最大的成分,且蛋白与多糖在好氧颗粒污泥与其在絮状活性污泥中的比值范围分别为3.22~5.80和1.68~2.63。
Abstract:
EPS was the main components of microbial aggregates, and the extraction and analysis of EPS for aerobic granular sludge was very useful for studying this novel microbial aggregate. Five methods including heating, ultrasonication, high-speed centrifigation, EDTA, and NaOH plus heating were employed for extracting EPS of aerobic granular sludge, and the key components of EPS extracted were analyzed. The experimental results reveal that the hemogenization was a necessary step for EPS extraction of granular sludge. Through comparison, the ultrasonication and heating were fit for qualitative analysis and quantitative measurement, respectively. For the ultrasonication (35W, 4min), the TOC in EPS extraction of activated sludge and granular sludge was 105.3 mg/g VSS and 96.5 mg/g VSS, respectively. For heating(80 ℃,60min), TOC production was 142.6mg/g VSS and 153.2mg/g VSS, respectively. Analysis for EPS extracted using different methods shows that the proteins were the most important components in the sludge samples, and the ratios of protein to polysaccharides in granular sludge and floc sludge were 3.22~5.80 and 1.68~2.63, respectively.
