蓝藻定向发酵产丁酸的条件研究

摘要: 以4种有机酸(乳酸、乙酸、丙酸、丁酸)对厌氧颗粒污泥进行胁迫处理,将胁迫后的污泥接种太湖蓝藻进行发酵产丁酸的实验,考察单一有机酸及混合有机酸胁迫对提高蓝藻厌氧发酵产丁酸的影响。结果表明,采用乳酸浓度2 g/L、乙酸浓度6 g/L、丙酸浓度5 g/L和丁酸浓度12 g/L的混合有机酸胁迫污泥后产丁酸效果最佳,获得的最大丁酸产量为10 252 mg/L,比空白提高了139%。实验中还对厌氧消化过程中蛋白酶活性的变化进行了测定,结果表明通过一定浓度的有机酸胁迫污泥,可以提高微生物的耐酸能力,进而提高蛋白酶的活性,最终达到提高丁酸产量的目的。Abstract: Four kinds of organic acids(lactic acid, acetic acid, propionic acid, butyric acid) stress were adopted in this experiment to pretreat the anaerobic granular sludge, then blue algae were inoculated with stressed sludge to produce butyric in order to evaluate the effect of stress on increasing butyric production. The results indicated that the tolerance of acid in sludge could be improved, thereby increasing butyric yield. The results show that the mixed organic acids which contains 2 g/L lactic acid, 6 g/L acetic acid, 5 g/L propionic acid and 12 g/L butyric acid has the best effect, the maximum butyric yield could reach 10 252 mg/L, which improved by 139% compared to control. The assay of protease activity during the fermentation in our study indicated that the appropriate concentration of organic acid stress could enhance the acid tolerance response of anaerobic sludge, improve enzyme activities, and ultimately raise the butyric yield.
