17β-雌二醇对西伯利亚鲟幼鱼的雌激素效应

摘要: 为了研究以及评估在利用17β-雌二醇(estradiol-17β,E2)混饲投喂进行人工诱导西伯利亚鲟(Acipenser baerii)性别逆转过程中,外源雌激素处理对西伯利亚鲟幼鱼的雌激素效应以及激素残留风险。实验选取健康的2月龄西伯利亚鲟840尾,设置E2在饲料中的投喂剂量为1 mg·kg-1(C组)和10 mg·kg-1(D组)的处理组,以及对照组(B组)和空白组(A组)进行实验,每组3个平行,每个平行70尾幼鱼,日投饵率为5%,实验持续90 d。利用全自动生化分析仪以及化学发光免疫分析仪分别测定雌激素处理前后血清磷以及E2含量变化;并采用半定量RT-PCR方法,分别检测卵黄蛋白原Vtg基因在不同E2处理组的鱼肝脏组织中的表达情况。研究结果表明:添加E2的实验组中,西伯利亚鲟血清中E2浓度显著升高(p < 0.05),停止E2处理2月后血清中E2浓度返回正常水平;Vtg基因在A组和B组未见表达,而在E2处理组被显著表达,且D组中Vtg/β-actin值为1.22±0.02,极显著(p < 0.01)高于C组的0.11±0.00。

Abstract: To study the estrogenic effect of estradiol-17β (E2) on the Siberian sturgeon (Acipenser baerii) and risk of estrogenic residue, 840 healthy Siberian sturgeons of two months old were randomly stocked into 12 tanks with 70 individuals per tank and fed with 4 kinds of diets containing various dosages of E2 (0, 1, 10 mg·kg-1) and equal ethanol. The E2 containing diet was administrated at 5% of the body weight per day for 90 days. The concentration changes of phosphorus and E2 in serum were determined by biochemistry analyzer and chemiluminescence immunoassay, and the Vtg (vitellogenin) gene expression in liver tissue was detected by RT-PCR. Results showed that the concentrations of E2 in serum in the treated groups were significantly higher than that in control and blank groups, and the concentration of E2 in serum regressed to normal levels after E2 was removed from the diet for 2 months. The Vtg gene signal was not detec-ted in liver tissue of Siberian sturgeon in the control and blank groups, but the Vtg gene was strongly expressed in the liver in the groups treated with E2. The ratio of Vtg/β-actin reached the value of 1.22 ±0.02 in the 10 mg·kg-1(E2/diet) treated groups, which was significantly higher than 0.11 ±0.00 in the 1 mg·kg-1 group.
