絮凝剂产生菌B-7的培养条件优化及生长动力学研究
Optimization of culture conditions for bioflocculantproducing bacterium B-7 and its growth kinetics

摘要: 从成都市土壤中筛选分离了1株具有稳定高效的微生物絮凝剂产生菌B-7。考察了碳源、氮源、温度、培养时间、pH等多种因素对絮凝剂(MBF-7)絮凝效果的影响。实验结果表明,该菌株产絮凝剂的最佳培养条件为: 碳源为淀粉,氮源为硫酸铵,培养时间为72 h,初始pH为7.0,温度为30℃, 对0.4%高岭土悬浊液的絮凝率达到91.3%。采用考马斯亮蓝、苯酚硫酸法、紫外扫描和红外光谱扫描分析絮凝剂MBF-7的成分, 测定结果表明, 絮凝剂MBF-7的主要成分为多糖类物质(质量分数为85.73%)。根据Logistic方程,得到的产絮菌B-7的生长动力学模型与实验数据能较好地拟合, 基本上反映了B-7菌体生长的动力学特征。Abstract: A bioflocculant-producing bacterium, named B-7, was isolated from soils of Chengdu city. The factors relevant to the culture conditions, such as the carbon source, the nitrogen source, temperature, culture time and pH were investigated and optimized. The results showed that under the following optimum conditions: starch as carbon source, ammonium sulfate as nitrogen source, initial pH of 7.0, temperature of 30℃ and incubation time of 72 h, the highest flocculation rate of 0.4% kaolin suspension was 91.3%. Meanwhile, the bioflocculan MBF-7 was mainly composed of polysaccharide and the total content was 73.48% (w/w) by the phenol-sulfuric acid method, coomassie blue reaction and ultraviolet spectroscope methods. According to the Logistic equation, the model and the experiment data were fit well and the model appeared to provide a reasonable description for growth kinetics of bacterial strain B-7.
