一株耐铜细菌的鉴定及富集特性的研究
Identification of a copper-resistant bacteria and its accumulation ability

摘要: 从铜矿污染区的土壤中分离纯化得到一株耐铜菌株USTB-E,最高耐受浓度为560 mg/L,通过形态观察,生理生化鉴定及16S rDNA序列分析技术鉴定出此菌株为假单胞菌。该菌的最适生长温度为35℃,pH为6.0,渗透压为0.3%。为了研究该菌耐铜机理,实验对铜离子富集机理进行了动力学研究,研究结果表明,USTB-E菌对铜离子有富集功能,有80.7%分布在细胞膜及细胞壁上,19.3%分布在细胞质内。因此,该菌株能够耐受高铜浓度的机理是将铜离子固定于细胞膜及细胞壁上来减低铜离子毒害。Abstract: A copper-resistant strain USTB-E was isolated from heavy metal contaminated soils, its highest tolerating concentration was 560 mg/L. According to pHysiological biochemical experiments and the 16S rDNA sequence analysis, it was identified as Pseudomonas sp.The optimum growth conditions of USTB-E were temperature of 35℃, pH of 6.0 and NaCl concentration of 0.3%. The results showed that Cu2+ could be accumulated by USTB-E. The distribution of Cu2+ in cell was 80.7% in cytolemma and cytoderm, and 19.3% in cytoplasm.
